The Problem with PCR

Moving Beyond Thermal Cycling

Traditional PCR requires precise temperature fluctuations (thermal cycling) to "unzip" DNA and amplify targets. While accurate, the physical limitations of heating and cooling mean that even the fastest PCR platforms bottom out around 30-45 minutes.

Isothermal amplification solves the speed problem by maintaining a constant temperature and using enzymes to unzip the DNA. However, historically, this speed came at a cost: lower specificity and "non-specific amplification" where resources are wasted replicating the wrong targets.
